How about Incredible Heights?
Any level that involves the sky, mountains, skyscrapers, or anything else that goes high up, or just a simple level that goes up.

History, where you build the level around a certain Historic event. It can have an entire story about it or just a straight platformer with the characteristics of the historical event of your choice.
(and it could be good for both SM63 and LL)

ooh are we posting ideas? This seems fun. Better post a crap ton in here.
-Pit of trials: basically a battle your way through enemies to get to the shine sprite thing. no open terrain allowed (except toward the end when you actually get to the shine sprite/end portal)
-Minigame Madness: a game WITHIN A GAME???? Pretty much just use game mechanics to make something other than the usual platforming and enemy stomping stuff.
-SPAAAAAAACE: Simply make a space themed level.
-RPG: make something with a massive plot and stuff with NPCs.
-I like it HARDCORE: Make an extremely difficult level.
-Uphill Climb: Level must not exceed 40 tiles WIDE, but it can be as high vertically as necessary.
-Items Only: Make a level with no tiles in it whatsoever. For LL Planets are allowed but blocks (tiles grouped together into a single item) are not.
-Typical: Make a level using the default Size. For SM63 this is 50 x 30 and for LL this is 150 x 50
-Back to the future: Make a level set either in the future or about time travelling to the future.

I have an idea here, I'm sure many of these are already LDC themes, but I'll suggest anyway.
I believe there is at least one LDC for each tileset's theme already, those are the typical themes of Jungle, Snow, Desert, Grass, Beach, Underwater, Volcano, Castle, Horror, Ruins, Sky, Space, Cave, Pipe Maze, etc.
But when I think about settings in many other video games, there are many more settings that are pretty common, but not so in SM63 because of the lack of an appropriate tileset.
Like City, Future, Food, Toys, Music (nothing to do with alt-music like the 16th LDC), Carnival, Flesh (you know, levels inside giant creatures), Casino, Oriental, Sewers, or maybe even Wild Western, etc.
However, with creativity, I believe these are probably all possible, such as City/Future, which was already used as LDC themes/level themes.
Anyway, perhaps one of these can be an LDC theme if not already used, or what do you think about an LDC where you must avoid common SM63 themes and do any of these instead?

Wild West sounds... Interesting. It'd be pretty hard to work with, but the result would pay off really well.
Also guys, remember to check out which LDC themes have already been used. A lot of the themes you guys suggested have already been used in a way or another.

There are kinda, uh, 156 pages of suggestions, plus 4 more in the LL version of this thread (which shouldn't exist and these need to be merged or we just need an entirely new thread)
