I'm not a huge block fan, but I do try to think of in-game what the best item is. Without a doubt, Normal Fludd. Its all around versatility is unmatched

ROTATING AND MOVING BLOCKS!!

You can do almost anything with them. including custom enemies: rain, lightning, 2nd mario, rainbow goombas, etc.

I must agree with Moving Blocks. You can make so many things. I have made bosses, fire, rain, lightning, enemies, pixel art, etc.

I think about 90% of the people here agree that it's the moving/rotating block. I personally enjoy it, as well as other item. However, the default Hover F.L.U.D.D. is a nice item too. Heck, add butt-loads of blue coins and you can easily make mini-games out of collecting them to get to the end of a level. (I for one, have used this) Those two items are probably my favorites. But, really, ANY item is good IF used right. I mean, think of all the 3D illusions you can make from rotating platforms and triangles.
