Not if you make sure to consistently use Transitions to reduce lag. I find levels more enjoyable when they are not multi-part unless there is a clear reason why they need to be multi-part, so if you can, I would recommend keeping it all as one level. I seem to recall that four's 3rd LDC entry (which won the gold) had an horizontal length of 998 tiles and a somewhat large vertical length as well and there was not a major amount of lag as far as I can remember.
